Fu incoronato Faraone a Menfi nel 197 aC Questo evento fu immortalato dalla Stele di Rosetta che Champollion trascriverà 2000 anni dopo, consegnando la chiave della scrittura geroglifica. Tolomeo V sposò Cleopatra I, figlia di Antioco III, e lasciò due figli minori: Tolomeo VI e VIII.
