Anciennement SALIS-SAMADE, le régiment reçoit le numéro 64 pendant la Révolution, en 1791, mais c'est sous l'Ancien Régime en 1672 qu'il est créé. Il participe aux guerres de Hollande, de la Ligue d'Augsbourg, de la succession d'Espagne, Malplaquet en 1709, Denain en 1712, et de la sucession d'Autriche. A l'origine sous le règne de Louis XIV c'est un régiment suisse servant la monarchie française.
Les trentes grenadiers qui défendirent la Bastille le 1' juillet 1789 en faisaient partie.
Ce régiment est licencié en 1792, mais, quatre ans plus tard, la 64e demi-brigade d'infanterie de ligne est constituée à partir de quatorze bataillons de volontaires issus de plusieurs départements dont celui de la Loire-Inférieure.
A partir de la Révolution , on trouve le 6-4 dans l'armée d'Italie et dans les armées de Rome et de Naples.
Il participe,aux guerres de l'Empire : Ulm, Austerlitz, Iéna, Espagne. En Espagne de 1808 à 1814, il passe en Algérie de 1841 à 1848, participant à la prise de la Smala d'Abd-El-Kader.
Puis ce sont, les campagnes d'Empire, armées d'Orient de 1855 à 1856, armée d'Italie en 1859 et la guerre de 1870, et le 6-4 retrouve l'Algérie de 1881 à 1882.
La 64e demi-brigade devient en 1863 le 64e Régiment d'infanterie de ligne. Y servirent les Maréchaux Bugeaud et Saint Arnaul.
