Tolomeo VI era il figlio maggiore di Tolomeo V e Cleopatra I Thea. La reggenza della madre durò fino al 176 aC Regnò insieme al fratello Tolomeo VIII dal 170 al 164 aC Sposò la sorella Cleopatra II dalla quale ebbe due figlie e Tolomeo VII. Morì per le ferite mentre combatteva contro Alessandro I Balas.
